Helicobacter pylori is the most important cause of gastritis, peptic ulcers and the development of gastric cancer. The chronic active inflammation is dominated by neutrophils, macrophages, lymphocytes and plasma cells. Several interleukins (IL-8, IL-10 and IFN-gamma) are involved in the inflammatory process in the gastric mucosa. The aim of this study was to investigate the gastric inflammation in patients with functional dyspepsia. Fifty-three consecutive patients were included and antral biopsies were obtained for histology, culture and immunohistochemistry. The sections were examined for the interleukins IL-4, IL-6, IL-8, IL-10 and IFN-gamma as well as for the cell markers CD4, CD8, CD14, Cd19, CD25 and CD30. Only CD4 and CD19 were significantly increased in patients with increased gastric inflammation and increased density of H. pylori. However, several of the examined markers (IFN-gamma, IL-8, IL-10 and CD14) showed a non-significant trend to be increased in patients with extensive gastric inflammation and high density of H. pylori. Therefore, an arbitrary index (IM11) for all the 11 immunological markers was made as an average value for each of the four morphological groups. For the four morphologically different groups of patients the values were 0.49, 0.77, 0.86 and 1.25, respectively. Significant increases in the index from none to moderate antral inflammation as well as the density of H. pylori were found (p<0.001). By using an index of inflammatory markers trends can be summarized and thereby significant which may be of importance when gastric inflammation is investigated in children and patients with functional dyspepsia.  相似文献

Insights into bacterium-host interactions and genome evolution can emerge from comparisons among related species. Here we studied Helicobacter acinonychis (formerly H. acinonyx), a species closely related to the human gastric pathogen Helicobacter pylori. Two groups of strains were identified by randomly amplified polymorphic DNA fingerprinting and gene sequencing: one group from six cheetahs in a U.S. zoo and two lions in a European circus, and the other group from a tiger and a lion-tiger hybrid in the same circus. PCR and DNA sequencing showed that each strain lacked the cag pathogenicity island and contained a degenerate vacuolating cytotoxin (vacA) gene. Analyses of nine other genes (glmM, recA, hp519, glr, cysS, ppa, flaB, flaA, and atpA) revealed a approximately 2% base substitution difference, on average, between the two H. acinonychis groups and a approximately 8% difference between these genes and their homologs in H. pylori reference strains such as 26695. H. acinonychis derivatives that could chronically infect mice were selected and were found to be capable of persistent mixed infection with certain H. pylori strains. Several variants, due variously to recombination or new mutation, were found after 2 months of mixed infection. H. acinonychis ' modest genetic distance from H. pylori, its ability to infect mice, and its ability to coexist and recombine with certain H. pylori strains in vivo should be useful in studies of Helicobacter infection and virulence mechanisms and studies of genome evolution.  相似文献

We developed a microarray hybridization-based method, 'comparative genome sequencing' (CGS), to find mutations in bacterial genomes and used it to study metronidazole resistance in H. pylori. CGS identified mutations in several genes, most likely affecting metronidazole activation, and produced no false positives in analysis of three megabases. We conclude that CGS identifies mutations in bacterial genomes efficiently, should enrich understanding of systems biology and genome evolution, and help track pathogens during outbreaks.  相似文献

The mitochondrial respiratory parameters were measured in situ, i.e. in saponin-skinned rabbit cardiac fibers and in fibers treated with saponin + collagenase. It was found that the decrease of maximal ADP-stimulated respiration rate of saponin-skinned fibers with pyruvate + malate under the conditions of total ischemia (0.5–1.5 h) is less pronounced as compared to isolated mitochondria. Maximal succinate oxidation rate (+ADP), however, was not different from control (1 h ischemia) but it exceeded the control level when measured in the medium supplemented with cytochrome c. It was also demonstrated that treatment of fibers with collagenase alone or in combination with saponin significantly (almost 2 fold) enhanced the maximal ADP-stimulated respiration rate if compared with saponin-skinned fibers. The data obtained suggest that mitochondrial respiration in saponin-skinned rabit cardiac fibers is not completely revealed, most probably, due to insufficient permeabilization of sarcolemma by saponin and, thus, inadequate accessibility of mitochondria to exogenous substrates, ADP in particular. The assessment of risks associated with non-indigenous species implies a detailed knowledge of their taxonomical composition and distribution within a certain region. The northern branch of the central European ‘invasion corridor’—a series of canals connecting different watersheds—has formed an important migratory route for Ponto-Caspian fish (i.e. fish from the Black Sea, the Sea of Azov and the Caspian Sea). However, the current status of non-indigenous fish species in this region is very scarce. This article presents a comprehensive overview of recent distribution data of non-indigenous fish species in the northern branch of the central European invasion corridor. Here, extensive data are integrated based on studies performed during 2003–2014 comprising reliable published and unpublished records from the last 12 years. Altogether, ten non-indigenous fish species were currently found in the northern branch of the central European invasion corridor, constituting 19 % of its freshwater fish diversity. Three species, including the Prussian carp (Carassius gibelio), the round goby (Neogobius melanostomus), and the Chinese sleeper (Perccottus glenii), are considered invasive species. Eight species may potentially invade this region in the near future. A comparison of the history of non-indigenous fish species introduction in the inland waters of the northern branch and other countries of the central European invasion corridor revealed similar introduction trends. Potential expansion of non-indigenous fish species across the central European invasion corridor has international implications that require awareness, cooperation, and government support from each individual country. Disclosure of recently operating vectors for non-indigenous fish introductions within the central European invasion corridor will help predict and prevent their further spread and establishment in this region.  相似文献

The rare microscopic red alga Galdieria phlegrea (Cyanidiohyceae, Rhodophyta) has been discovered in the extremely acid Tinto River in Spain and this occurrence is here related to previous knowledge about the distribution and ecology of this enigmatic alga. The taxonomic affiliation of the new isolate of G. phlegrea was revealed by reconstructing the phylogeny of plastid‐encoded rbcL. According to this phylogeny, the Tinto River alga is closely related to other G. phlegrea strains originating from extreme habitats in Czechia, Italy and Turkey, suggesting a wider distribution and higher ecological versatility than previously thought. The results suggest that G. phlegrea, and then possibly also other cyanidiophycean algae, are not as restricted to strongly acidic and hot microhabitats as previously believed, which, in turn, may indicate that they may commonly have been overlooked and possibly are much more widespread than is currently believed.  相似文献

The germline polymorphisms in signal-inducing proliferation-associated protein 1 (SIPA1) and ribosomal RNR processing 1B (RRP1B) might be involved in breast cancer metastasis. The aim of this study was to analyze how SIPA1 and RRP1B polymorphisms contribute to breast cancer phenotype, lymph node status and survival. A group of 100 young, I–II stage breast cancer patients were analyzed for SIPA1 and RRP1B polymorphisms with PCR-RFLP assay. SIPA1 c.2760G?>?A, c.545C?>?T and RRP1B c.436T?>?C polymorphisms were associated with lymph node status, survival and tumor grade, respectively. Our results suggest that SIPA1 and RRP1B germline polymorphisms are important for breast cancer prognosis.  相似文献
